Best selling drugs

Erythromycins from Streptomyces erytkreua strains are probably the structurally most complex of the best selling drugs. Erythromycin A costs only about 5 DM per gram. The two principal erythromycins A and B differ only with respect to hydroxylation at C-12. 

Patents on current best-selling drugs are expiring. It has been estimated that the top 100 products in the marketplace will all come off patent (that is, the basic patents will expire) in the period between 1973 and 1990. 

Esomeprazole is the S-isomer of omeprazole (Prilosec), which is a mixture of the S- and R-isomers. Prilosec, for many years a best selling drug, is 5-methoxy-2-[[(4-methoxy-3,5-dimethyl-2-pyridinyl)methyl]sulfinyl]-IH-benzimidazole. Its empirical formula is (Ci7Hi8N303S)2Mg-3H20 with a molecular weight of 767.2 as a trihydrate and 713.1 on an anhydrous basis. 

Lipitor is currently one of the best selling drugs for lowering cholesterol. Searching in WDI for Lipitor produces the page shown in Figure 10.1 [46]. The preferred name is Atorvastatin Calcium . Statins are compounds that inhibit HMG-CoA reductase, the rate-limiting enzyme in cholesterol biosynthesis, and they are the world s best-selling drugs and are used for lowering cholesterol. Statins are well studied and are believed to be quite safe. Because they reduce the levels of cholesterol, the precursor of the bile acids, statins may be the ideal drugs to use for BA-loweiing in these GI tract diseases.

A THR analysis of 293 marketed drugs demonstrated that over 60% were selective (THR <5% Figure 12.3). This group included 22 antipsychotics, which were all promiscuous. A subset of 132 of the most prescribed and best selling drugs, excluding any withdrawn drugs or antipsychotics, had only 5% promiscuous but 73%...

Fenfluramine, a racemic chemical more closely related to amphetamine than to serotonin, affects brain levels of serotonin and its metabolites at relatively low doses. It was introduced as an anorexigenic drug almost 20 years ago. Its d- isomer, J-fenfluramine (dexfenfluramine, Redux) was about twice as potent as fenfluramine. After several controlled clinical trials demonstrated that dexfenfluramine could help patients maintain weight loss for at least a year, it was quickly introduced into the market in the USA and became a best-selling drug overnight.

Of the top 10 best selling drugs of 2010, GlaxoSmithKline s Advair, an inhaled asthma medication, was omitted from question 6. Why was this drug left out of the Lipinski analysis of question 6 ...

Research groups in Japan [1] have screened over 8000 microbial extracts for their ability to produce an inhibitor of sterol synthesis in vitro. These studies led to the isolation of mevastatin from cultures of Penicillium citrinum. Mevastatin (Fig. 4.2) served as the lead-molecule for seven statins which currently are among the best-selling drugs.

A similar PCA analysis of 595 natural products from the Merck sample collection, 137 small molecules from the top 200 best selling drugs in 2006 and the 65 000 sample combinatorial library from ChemBridge (one of the libraries used by Feher and Schmidt in their analysis) showed a similar plot and provided similar conclusions (Figure 2.5).
