Shake-Flask Experiments with T ferrooxidans. A limited number of experiments were conducted using T.ferrooxidans to determine the influence of process variables on the rate and extent of pyritic sulfur release from the coal samples in shake-flask experiments using a mechanical shaker. No attempt was made to optimize the mineral salts medium composition since the influence of NtfJ, N/P molar ratio and the nitrogen requirements for the growth of T. ferrooxidans have been thoroughly investigated by other workers (4,7,8,10). A mineral salts medium with the composition described earlier in the Microbial Procedures section was used in all the experiments with T. ferrooxidans. 

Normally, for the partial oxidation processes, only a high-temperature shift conversion is used. This results in a carbon monoxide content of the gas after shift conversion of 3 vol % or sometimes somewhat higher. Liquid nitrogen wash [754] - [756] delivers a gas to the synthesis loop that is free of all impurities, including inert gases and is also the means for adding some or all of the nitrogen required for synthesis.

Air, methane, and any further hydrogen available are then added to the exit gas stream from the steam reformer in the appropriate proportions to provide all of the eventual nitrogen requirement for ammonia synthesis (Eq. 11.20). 

Unlike most carbohydrate and lipid, protein contains nitrogen required for incorporation into a wide range of compounds (Table 22-4). Although the synthesis of proteins fixes nitrogen into those proteins, they are synthesized in direct response to specific needs. Dietary protein nitrogen is used for the synthesis of nitrogen-containing compounds in response to specific hormones suchas insulin. 

Of the 20 amino acids in proteins, the body can readily synthesize eight if an appropriate nitrogen source is available. Two others can be synthesized from other amino acids of the diet tyrosine from phenylalanine and cysteine from methionine. The rest must be provided in the diet (Chapter 17), since the body can synthesize none or an insufficient amount. The dietary requirement depends on several factors. Beside essential amino acids, the diet should provide the nitrogen required for synthesis of the nonessential amino acids. 

Nitrogen requirements for the patient with liver failure are not unlike those of normal subjects, but intolerance to protein is common, and protein restriction has been used successfully as part of the therapy. A dilemma arises when the diet becomes so restrictive that malnutrition results, and the patient becomes susceptible to infection and other comphcations. Overzealous use of protein to correct nutritional deficits invariably results in hepatic encephalopathy.
