Nested Polymerase Chain Reaction. For the first round of amplification, 20 to 50 pi of cell lysate, 2 pi of DNA-RNA or 10 pi DNA-cDNA is amplified for 30-35 cycles in an 100//1-reaction volume. As products from the first amplification are usually not visible on the gel with ethidium bromide staining, 5 pi of the first amplification products is then used to seed a second 100 pi reaction and amplified for another 35 cycles.
