The use of tissue slices is a standard technique in all aspects of metabolic biochemistry. The tissue is cut into slices, sufficiently thin to allow adequate rates of diffusion in and out of the tissue. The slices are submerged in physiological saline to which substrates or other compounds may be added.

Thiamin was the first of the vitamins to be demonstrated to have a clearly defined metabolic function as a coenzyme indeed, the studies of Peters group in the 1920s and 1930s laid the foundations not only of nutritional biochemistry but also of modern metabolic biochemistry and neurochemistry. Despite this, the mechanism by which thiamin deficiency results in central and peripheral nervous system lesions remains unclear in addition to its established coenzyme role, thiamin regulates the activity of a chloride transporter in nerve cells.

To examine the cell and its biochemistry it is necessary to interfere in some way with its normal functioning, and interpreting the results of this interference in terms appropriate to the reality of the cell in situ, rather than the experimentally reduced artefact, requires the highest degree of critical awareness on the part of the experimenter. One must never forget that the substrate, enzyme, or cell is never really an isolate. Our experiments have to make it so to study it, but to interpret them, we must consider always the whole system. Metabolic biochemistry is fun, but it has its dangers ...
