Lung lobes

The ventilated and perfused human lung lobe was used as described by Linder and co-workers [74], A twofold difference in the appearance of drug and metabolites in the perfusate was found for the two formulations. Small fractions of the applied dose of BDP were immediately detectable in the perfusate and the amount of the major metabolite, beclomethasone-17-propionate (17-BMP), increased over the experimental period. These observations were similar to the clinical observations that BDP is detected rapidly in the plasma after inhalation and that the appearance of the active metabolite 17-BMP occurs rapidly. The kinetic differences between the formulations were explained on the basis of particle size effects with the conclusion that the discriminatory value of this system to examine the lung pharmacokinetics of inhaled medicines in the absence of systemic effects such as hepatic metabolism was apparent. 

Remove the lung lobes by sectioning the bronchial tubes, weigh together.

The lung lobes are shredded against each other using 200 strokes of two pairs of closed fine curved forceps. A dark background is advantageous, and tissue can also be pulled free of remaining airways. 

The systemic bioavailability of recombinant human granulocyte colony stimulating factor (rhG-CSF) based on the amount reaching the lung lobes following intratracheal instillation to the hamster was estimated to be 62% relative to intracardiac administration. Remarkably, about 20% of the absorbable dose of this 18.8 kDa protein was present in serum within 6 min... 

Most monkeys had similar microscopic pulmonary lesions (Figure 31-1). The most obvious lesion was marked multifocal-to-coalescing interstitial pulmonary edema involving multiple lung lobes. Peribronchovascular connective tissue spaces were distended by pale, eosinophilic, homogeneous, proteinaceous material (edema) ...

Another unique movement is shown at head side measurement. The displacement of Y-direction movement initially started to move downward, but then moved towards its initial heighL and finally reached higher than initial location. This is observed to be because of existence of lung lobes. Each lobe consists of its own independent bronchioles and blood vessels. Since measured points were crossing different boundaries of lobes, this kind of displacement behavior are seen due to independent lethargy process of each lobes. Same behavior of displacement is observed to hip side experiment of measure point number ,(D, and of Y-direction. 

Direct application of the tgAAVCF vector to the nasal epithelium and a single lung lobe revealed no troubling safety issues, but it also did not provide evidence of efficient gene transfer or expression at the sites of administration. However, evaluation of aerosolized vector delivery to CF patients provided data that indicated that AAV vectors could produce gene transfer in the lung.
