Lugol’s iodine

Lugol s iodine 1% iodine and 2% potassium iodide in distilled water.

Tissues are fixed with buffered formalin or Kryofix and embedded in paraffin (Boon et al., 1991). Sections (5 xm thick) are transferred to a glass slide, deparaffinized, rehydrated, and washed in running tap water for 10 min. They are treated with Lugol s iodine for 5 min and rinsed briefly in tap water. Following destaining with 2% aqueous sodium thiosulfate for 10-15 sec, the sections are washed in running tap water for 10 min. 

Standard hydrographic data (temperature, salinity, fluorescence and inorganic nutrients) were generally recorded continuously. Chlorophyll a concentrations of discrete samples were determined fluorometrically. Water samples were preserved with Lugol s iodine solution and 0.5% neutralised formaldehyde for onshore identification and enumeration of the phytoplankton using an inverted microscope.

Place sections in Lugol s iodine for 5 min, then in 2.5% sodium thiosulfate for 30 s if IGSS method is to be used.

Iodine was discovered by Bernard Courtois in 1811 and soon used in the treatment of water. It was the introduction of Lugol s iodine (or Lugol s solution) in 1829 that offered a more readily available form for immediate disinfection of drinking water. Iodine has been used as a disinfectant in military applications since World War I (White, 1999). In 1952, the current form of globafine tablets was first integrated into field use for water treatment by the U.S. Army and the tablets were widely distributed to those operating in austere environments (Powers, 1991). Some dyes have a specific interaction with certain polysaccharides e.g. Congo red interacts with (l->4) P-D, (l->3) P-D and (l->3)(l->4) p-D glucans and (l->4) p-D xylans (11, 12). Upon flooding the plate with the dye-solution, undegraded polysaccharide will be stained by the dye whereas degradation zones will remain uncoloured. Congo red was also applied directly in the medium with CMC (13). Another classical example is the blue coloured complex that is formed between intact amylose and Lugol s iodine solution.
