F. Enzyme inhibition Enzymatic reactions can be surveyed on a TLC plate, and the end products detected. For example, cholinesterase-like enzymes in nervous tissue will catalyze the substrate indoxyl acetate to indoxol and acetic acid. In air, indoxol is converted to indigo blue, which can be visualized on a TLC plate. Some pesticides and insecticides inactivate enzymes associated with animal nervous tissue. The presence of minute amounts of such substances may interfere with the indoxyl acetate substrate reaction producing colorless spots on a blue background. According to another paper, methamidophos and phosphorus-containing insecticides were detected by beef liver esterases in Tris buffer using indoxyl acetate, K3pe(CN)6, and K4Fe(CN)g. The insecticides gave white spots on a blue background with a detection limit of 15 ng/spot (162). 

Apart from a report of the rather surprising use of indoxyl acetate as a fiuorogenic substrate (Guilbault et al., 1967 Rhodes el al., 1970), conventional hyaluronidase assays employ giycosaminoglycans for this purpose. Procedures have also been described for the visualization (Abrahamson and Friedman, 1967) and histochemical location (Davost, 1965) of hyaluronidases. 

In ester synthesis and exchange reactions, as well as in hydrolysis re tions induced by PEG-lipase in hydrophobic media, the existence of a trace amount of water in the reaction system was most important in terms of the reactions proceeding. Matsushima et al. [67] carried out a kinetics study of PEG-lipase in transparent benzene solution to estimate the value of water, one of the substrates of lipase in the ester hydrolytic reaction. Indoxyl acetate was hydrolyzed by PEG-lipase to form acetic acid and 3-hydroxyindole, which was photometrically determined. A double-reciprocal plot of the velocity of the indoxyl acetate hydrolysis against water concentration at a given concentration of indoxyl acetate indicated that the hydrolysis took place as a double-displacement reaction (ping-pong reaction). The apparent Michaelis-Menten constant of water and the maximum velocity were calculated to be = 7 X 10 M and Vmax = 4700 xmol/min/mg of protein, respectively.
