Today it has become clear that the effect of trace elements in living systems, in food, and in the environment depends on the chemical form in which the element enters the system and the final form in which it is present. The form, or species, clearly governs its biochemical and geochemical behaviour. lUPAC (the International Union for Pure and Applied Chemistry) has recently set guidelines for terms related to chemical speciation of trace elements (Templeton et al. 2000). Speciation, or the analytical activity of measuring the chemical species, is a relatively new scientific field. The procedures usually consist of two consecutive steps (i) the separation of the species, and (2) their measurement An evident handicap in speciation analysis is that the concentration of the individual species is far lower than the total elemental concentration so that an enrichment step is indispensable in many cases. Such a proliferation of steps in analytical procedure not only increases the danger of losses due to incomplete recovery, chemical instability of the species and adsorption to laboratory ware, but may also enhance the risk of contamination from reagents and equipment. [Pg.75]

To analyse the solvent extracts a lOpL aliquot of the concentrated extract was injected into a Finnigan 4000 gas chromatograph-mass spectrometer coupled to a 6110 data system. A 3% OV-17 provided the best separation of the detectable PAHs. Quantitative estimations of the detectable O-PAHs in Ottawa drinking water were obtained by comparison of the areas of the two characteristic ion peaks (Tablesl6.2 and 16.3) in the mass chromatograms of the reference standard and the field sample, respectively. No corrections were made for incomplete recovery. Of the 50 PAHs in the standard used by Benoit et al. [119], 38 are detected in at least one of the drinking water samples tested. [Pg.413]

An important distinction must be made between the humoral response to a pure, capsular polysaccharide, and to the same polysaccharide when it is an integral part of the bacterium. Thus, the immunity received on recovery from infection by encapsulated bacteria, in terms of the polysaccharide antigen, differs from that generated by purposeful immunization with purified capsular-polysaccharide vaccines. Fortunately, with the exception of infants, the polysaccharide vaccines still stimulate protective-antibody levels in humans, despite these differences. In infants, due to the immature nature of their immune systems, these polysaccharide vaccines are of only marginal benefit.7 Some insights into the nature of these different responses in humans can be found in studies on the cellular basis of the immune response to polysaccharides.
