Immunoglobulins affinity purification

Our experimental system is based on HFN and antibodies from the immunoglobulin G fraction before and after affinity purification. The immunoglobulin G fraction from an immunized rabbit contains not only HFN specific antibodies but also antibodies to other antigens. The HFN specific antibodies can be purified by affinity chromatography. The surface concentration of HFN interacting with surface immobilized antibodies is thus dependent on the purity of the antibodies (figs. 2 and 3).

Other common impurities, such as immunoglobulins and protein A, result from the immunoaffinity purification of recombinant proteins or MAbs.16 If affinity chromatography is used to purify an antigen, then an ELISA can be used to detect contaminating levels of MAbs leached from the column. An assay for the antibody needs to detect the antibody in the presence and absence of its specific antigen. 

Staphylococcal protein A (SPA), with its immunoglobulin binding ability, has been used as an affinity tail for the purification of a human insulin growth factor-1 (lGF-1) fusion. The insertion of an acid-labile Asp-Pro cleavage site at the fusion point allowed the separation of the protein A moiety from lGF-1 (Nilsson et al., 1985).
