Church [39] proposed the use of the ratio of the expected values of Martin s and Feret s diameters as a shape factor for a population of elliptical particles. Cole [40] introduced an image-analyzing computer (the Quantimet 720) to compare longest chord, perimeter and area for a large number of particles. Other parameters have been proposed by Pahl el. al [7,41], Beddow [42] and Laird [29]. 

Visual comparison of the holographically recorded radiation pattern of a particle with Mie theory has also been used for particle sizing [71]. Holography has also been used to locate sub-micron particles in a 3-dimensional volume [72] and, in conjunction with an image analyzing computer, to size the droplets in sprays [73]. 

One of the earliest imaging systems to be used in conjunction with microscopy was the Quantimet Model 720 system, an automated image analyzer [102,103]. The pattern recognition modules of the image-analyzing computer allowed features to be classified by orientation-independent size criteria, such as area and perimeter. By application of a wide range of complex shape, size, and density criteria, it was also possible to separate features by shape. 

Image analyzing computers are now routinely applied to the measurement of structures observed in microscope images. Quantitative microscopy involves several steps which all must be considered as part of the analysis. The sample must be prepared and the actual microscopy performed. Crystal structure of a protein molecule can also be determined by x-ray crystallography. Purified protein is crystallized either by batch methods or vapor diffusion. X-rays are directed at a crystal of protein. The rays are scattered depending on the electron densities in different positions of a protein. Images are translated onto electron density maps and then analyzed computationally to construct a model of the protein. It is especially important for structure-based drug designs. 

Our algorithms were verified by analyzing images from computer generated three-dimensional structures. 

The absolute reference method of image analysis of fiber bundle cross sections [572-574], wherein an image analysis computer system is used to automatically measure the area and perimeter of several hundred fiber sections and statistically analyzed to measure the average 0 and perimeter.

The Cambridge Instruments Quantlmet 900 Image Analyzer was used to analyze the radiation tracks on exposed CR-39 polymer. This system features a computer controlled stage and focus system. We have shown that commercial CR-39 monomer samples can be purified by column chromatography. Polymerization with benzoyl peroxide as Initiator gives highly crossllnked polycarbonate samples suitable for use as high energy radiation dosimeters. A method has been developed to characterize the polymer and to count radiation tracts with a computer controlled Quantlmet Image analyzer.
