The next abstraction level of reaction retrieval is a so-called reaction substructure search in which both query structures arc considered as substructures. In the case of a reaction substructure search, no hydrogen atoms arc added internally during the execution of the search. A good example of a surface-modified lens is the Sola/Bames-Hind Hydrocurve Flite lens, introduced in 1986. The material for the commercial Hydrocurve lens, bufilcon A [56030-52-5] contains methacrylic acid and has a high affinity for protein and subsequent deposition. The surface of the Flite lens was chemically modified with the addition of diazomethane (190) to reduce the surface charge. The polypeptide chain of the lac repressor subunit is arranged in four domains (Figure 8.21) an N-terminal DNA-hinding domain with a helix-turn-helix motif, a hinge helix which binds to the minor groove of DNA, a large core domain which binds the corepressor and has a structure very similar to the periplasmic arablnose-binding protein described in Chapter 4, and finally a C-terminal a helix which is involved in tetramerization. This a helix is absent in the PurR subunit structure otherwise their structures are very similar. [Pg.144]

The light-harvesting complex LHl is directly associated with the reaction center in purple bacteria and is therefore referred to as the core or inner antenna, whereas LH2 is known as the peripheral antenna. Both are huilt up from hydrophohic a and p polypeptides of similar size and with low hut significant sequence similarity. The two histidines that hind to chlorophyll with absorption maxima at 850 nm in the periplasmic ring of LH2 are also present in LHl, but the sequence involved in binding the third chlorophyll in LH2 is quite different in LHl. Not surprisingly, the chlorophyll molecules of the periplasmic ring are present in LHl but the chlorophyll molecules with the 800 nm absorption maximum are absent. [Pg.242]
