Stem cells hematopoetic

Osteoclast differentiation Raw264, bone marrow macrophages, hematopoetic stem cells Rankl Comparison of response markers across models to identify common mechanisms [23]... [Pg.420]

The study of osteoclast differentiation is important for understanding potential new treatments for osteoporosis. Such therapies are typically explored in tissue culture models such as the Raw264 mouse monocytic cell line, which is capable of differentiation into functional multinuclear osteoclasts after treatment with the cytokine Rankl. Use of transformed cell lines raises the concern that results may not be extrapolated to normal tissue. To address this question, the transcriptional responses for Rankl treatment of the Raw264 cell line, and of two ex vivo primary cell systems (bone marrow macrophages, and hematopoetic stem cells) were compared using Affymetrix GeneChips [23]. The models proved to... [Pg.421]

A BMU is generated, and a cycle commences, when circulating hematopoetic stem cells from the marrow are recruited, fuse, and differentiate into multinucleate osteoclasts (Figure 25(a)). Enhanced by circulating hormones, osteoclasts resorb a portion of mineralized tissue a few micrometers in size, creating lacunae, or... [Pg.4026]

Bone marrow was harvested from adult male mice and then transplanted into Isogeneic female recipient mice that had been treated with a level of irradiation sufficient to destroy their own hematopoetic stem cells (see the figure). Although the dose of irradiation given was lethal to mice that did not receive a transplant, the majority of the recipient mice receiving the transplant survived. After 4 weeks, peripheral blood from the recipient mice was analyzed. The composition of the blood was normal with respect to all blood cell types. Every blood cell examined was determined to be positive for the presence of the Y chromosome. [Pg.932]

Hematopoiesis is the continual process of blood cell production and tbe development of the various cell lines. Blood cells are the progeny of a hemato-poetic stem cell. Through the process of hematopoiesis, the body is supplied with ample blood cells of aU lines. The hematopoetic stem cell can both replicate itself and differentiate into the mature cells found normally in the peripheral blood. The hematopoietic system includes the bone marrow, liver, spleen, lymph nodes, and thymus. Regulatory growth factors also play a role in hematopoiesis. [Pg.954]
