Glycoprotein ovomucoid

Proteins, amino acids bonded through peptide linkages to form macromolecular biopolymers, used as chiral stationary phases for hplc include bovine and human serum albumin, OL-acid glycoprotein, ovomucoid, avidin, and cellobiohydrolase. The bovine serum albumin column is marketed under the name Resolvosil and can be obtained from Phenomenex. The human serum albumin column can be obtained from Alltech Associates, Advanced Separation Technologies, Inc., and J. T. Baker. The a1-acid glycoprotein and cellobiohydrolase can be obtained from Advanced Separation Technologies, Inc. or J. T. Baker, Inc. [Pg.66]

In these copolymers, the saccharide block is a carbohydrate fraction (0Q or Op) of a glycoprotein (ovomucoid) and the peptide block is a poly(benzyl-L-glutamate) block or a poly(cinnamyl-L-glutamate) block. [Pg.149]

It is well known in biochemistry that many proteins, especially enzymes but also transport proteins such as albumin, show high enantioselectivities in their interactions with small chiral molecules. It is possible to bind proteins to silica and to obtain a valuable class of CSPs that is mainly suited for the separation of chiral drugs. Several protein phases are commercially available albumins, a-acid glycoprotein, ovomucoid, avidin, cellobiohydrolase I, and pepsin. These proteins are composed of simple proteins in combination with carbohydrates like mucopolysaccharides, which include hyaluronic acid and chondroitin sulphates. On hydrolysis, mucopolysaccharides yield more than 4% of amino-sugars, hexosamine and uronic acid e.g., ovomucoid from egg white. Soluble mucoproteins are neither readily denatured by heat nor easily precipitated by common protein precipitants like trichloroacetic acid or picric acid. Since ovomucoid is a glycoprotein the solutes that can be stereo-chemically resolved on the OVM CSP are similar to the basic, neutral, and acidic compounds separated on the AGP CSE The major differences between the two CSPs appears to be quantitative i.e the magnitude of the chiral separation), rather than qualitative.
