Glycol linker

Using the same versatile modular synthetic strategy, the same group developed biotinylated bi- (438) and tetra-antennary (439) mannosylated glycoconjugates to capture and detect E. coli cells, and compared the relative capturing ability of these molecules to commercial polyclonal antibodies (Fig. 47).318 Instead of aliphatic spacers, tetraethylene glycol linkers were used to diminish nonspecific binding and to impart flexibility for a better fit in the active sites.

Identification of Molecular Target To identify the molecular target, Chen et al. synthesized three myriocin derivatives, which bear a pentaethylene glycol linker at (1) the aliphatic extremity of myriocin (C20-linker, 2), (2) a primary amine (N-linker, 3), and (3) a carboxyl terminal (Cl-linker, 4) [6]. Carbohydration and polyethylene glycol linkers were combined with chemoselective conjugation to optimize the pharmacokinetics of the peptides investigated. Oxime formation was successfully adopted for chemoselective F-labeling of an aminooxy-modified affibody (Namavari et al. 2008) and a suitable derivative of the hormone leptin (Flavell et al. 2008).
