All assay methods are based on the forward ALD-catalyzed reaction. Both photometric fixed-time and continuous-monitoring procedures have been developed. In the analytical approach on which all the commonly used procedures and kits are based, the ALD reaction is coupled with two other enzyme reactions. The Km for G-3-P is 1.1 x 10 M, for NAD it is 3.8 x lO M and for dihydroxyacetone it is 4.6 x lO M in phosphate buffer pH 7.0 and at 23.3°. Dihydroxyacetone phosphate and fructose-1,6-diphosphate are inhibitors.
