Platelet Adhesion from Washed Platelet Suspensions... 

Thrombosthenin is extracted from a concentrated suspension of washed blood platelets obtained by any of the described methods of isolation (Maupin, 1954b). Bettex-Galland and Liischer (1961), starting with 50 liters of freshly (collected citrated human blood isolated by differential centrifugation in the (told, from the buffy layers, 20 to 30 ml of a highly concentrated suspension of washed platelets. Since the isolation of thrombosthenin is based on its solubility properties, special ce,re must be taken to eliminate the leucocytes their content of deoxyribonuclcoproteids...

A similar series of experiments concerned a blend of a terpolymer of PVC with a cationic elastomer, containing or not ionically bound heparin. Perfusion with washed platelet suspensions, under conditions excluding thrombin generation from plasma, demonstrated the activation of platelets by heparin 3U), as reflected in a parallel increase in platelet deposition and RTG secretion (Table V). The perfusion system thus offers a simple means of in vitro screening of biomaterials for short term interactions with platelets and plasma proteins. 

To assess the effect that any air nuclei have on cellular acftiesion to silicone rubber, we first compared the platelet adhesion from blood and washed platelet suspensions when either of them were exposed to normally primed and to denucleation primed specimens of silicone rubber. 

The washed platelet suspensions were prepared from swine blood (9. 10) and the platelet concentrations in them were set at the normal value (2.5 x 10 platelets/mm ). As measured by the Lowry technique (ID. the total protein concentration was reduced to an aver of 0.018 mg/mL which is a very small fraction of the normal value.

Srivastava et al. showed that curcumin inhibited platelet aggregation induced by arachidonate, adrenaline, and collagen. Miscellaneous Agents - It has been reported that high concentrations of penicillin 6 are capable of inhibiting ADP, collagen and thrombin induced PA as well as the platelet release reaction in human PRP and in suspensions of washed platelets from rabbits or pigs.26... 

The fact that BN 52021 displaces the [3H]PAF from its receptor like unlabelled PAF in both human [120, 190] and rabbit [187] platelets further demonstrates its high specificity as an antagonist. The potent binding capacity of BN 52021 to PAF receptors is also evidenced by the fact that platelets pretreated with the antagonist and then washed do not respond to further stimulation by PAF [120, 190].
