Figure 11.1a [1] shows a schematic representation of a micropreparative thin-layer chromatogram obtained on a 0.5-mm Florisil (magnesium silicate) layer prewetted with benzene of a crude extract, i.e., containing coextracted plant oil obtained from Heracleum moelendorfi fruit. The initial band of extract was washed with benzene and then separated by continuous development with ethyl acetate in benzene [1]. As seen from the fraction analysis presented in Figure 11.1b, small quantities of pure bergapten and xanthotoxin can be isolated in this maimer.

In agreement with Steel s formulae [24], where HCT-116 cells show a doubling time of 18 2 hours with TGi 9 2, TGi 4 2 and TG2m 3 1 hours, cells were analyzed at 7, 16 and 24 hours for in vitro kinetics and up to 72 hours for cell loss studies. Unlabelled fraction analysis show edotecarin was more efficient than SN-38 to block cell proliferation of Gi and G2/M fractions during shorter treatment while at a lower concentration, while for SN-38 a higher concentration (Figure 8) was required. 

Once the sample preparation is complete, there are several approaches to the analysis of petroleum constituents in the water and soil (1) leachability or toxicity of the sample, (2) the amounts of total petroleum hydrocarbons in the sample, (3) petroleum group analysis, and (4) fractional analysis of the sample. These methods measure different petroleum constituents that might be present in petroleum-contaminated environmental media.

Retention times of the peaks are subject to the particular type of column. The acidic fraction from solid-phase extraction consists of phenolic acids such as cis-coutaric, trans-coutaric, and trans-caftaric acids. Isocratic elution is suitable because of the limited number of compounds found in the acidic fraction. Analysis of the acidic fraction is completed within 30 min. See Figure 11.3.1 for an HPLC chromatogram of the acidic polyphenolics isolated from Niagara grapes.

In more practical terms there will be SOPs to describe sample preparation, equipment set-up including programming, and an SOP for operation of the equipment. The documentation does not stop here since there will be defined instructions for fraction analysis, fraction reconciliation and recovery of product from the fractions. 

The effect of tacticity of the polymeric backbone on properties of POCT-4 has been searched for [26], Figure 4 shows curves for PMCS-4 with different backbone structure, obtained by the diffe-rential scanning calorimetry (DSC) method (curves 1 - 4). For atactic polymer the only transition, corresponded to Tg, is observed on curve 1, above which, in accordance with the data of X-ray dif-fraction analysis, the polymer is amorphous (two amorphous haloes at 20 = 8-11° and 20-35°).
