Fine chemicals drug development stages

This chapter will attempt to summarize the application of chiral auxiliaries at a scale important to the chiral fine chemical business. In addition, and as important, there are numerous cases where a chiral auxiliary has been used in the early stages of pharmaceutical development and the potential exists for this approach to be used in future work at larger scale. Last, there exists a limited number of cases where the auxiliary exists as part of an active drug. The known industrial applications, the potential applications, and the presence of auxiliaries in active drugs will all be addressed. [Pg.443]

As described previously, virtual screening and in silico design will accelerate the discovery of active lead compounds with new chemical scaffolds. The in silico prediction of physicochemical and ADME (absorption distribution metabolism elimination) properties, however, also are very critical for lead development. Actually, pharmacokinetics and toxicity have been identified as important causes of costly late-stage failures in drug development. The recently developed in silico approaches will increase model productivity in fine-tuned lead optimization to improve compound design and lead optimization. [Pg.126]
