Analytical separations may be classified in three ways by the physical state of the mobile phase and stationary phase by the method of contact between the mobile phase and stationary phase or by the chemical or physical mechanism responsible for separating the sample s constituents. The mobile phase is usually a liquid or a gas, and the stationary phase, when present, is a solid or a liquid film coated on a solid surface. Chromatographic techniques are often named by listing the type of mobile phase, followed by the type of stationary phase. Thus, in gas-liquid chromatography the mobile phase is a gas and the stationary phase is a liquid. Acoustic Wave Sensors. Another emerging physical transduction technique involves the use of acoustic waves to detect the accumulation of species in or on a chemically sensitive film. This technique originated with the use of quartz resonators excited into thickness-shear resonance to monitor vacuum deposition of metals (11). The device is operated in an oscillator configuration. Changes in resonant frequency are simply related to the areal mass density accumulated on the crystal face. These sensors, often referred to as quartz crystal microbalances (QCMs), have been coated with chemically sensitive films to produce gas and vapor detectors (12), and have been operated in solution as Hquid-phase microbalances (13). Solid phase extraction (SPE) is a very simple, rapid and reproducible cleanup technique that is now widely accepted as an alternative to the time-consuming liquid-liquid extractions. Additionally, SPE uses relatively small volumes of solvents, and is easy to automate. It is available in a number of different formats, including cartridges, disks, loose material, well plates or SPME using film-coated capillaries. SPE can be considered as an extraction technique when used for isolation and concentration or a cleanup technique when used to remove co-extractives from solvent extracts.
