Ferulic acid matrix

Despite the large number whole-cell MALDI protocols that have been tested a single approach has not yet been widely adopted. Experiments using a matrix of four levels of ferulic acid and four levels of moisture stress demonstrated that the combined action was additive under more stressful levels of the individual factors than in the previous tests. Duke et al. (23) tested the germination of lettuce seeds treated with phenolic acids (1 mM) at water potentials (D-mannitol) of 0, -0.2, -0.4, and -0.6 MPa. The combined action of low water potential and exposure to phenolic acids resulted in an additive detriment to germination, and the authors concluded from probit analysis that the mechanism of action from these sources was similar. Whatever their mechanisms, moisture stress and phenolic acids appear to work together in limiting growth of plants.

Both cutin and suberin contain phenolic components that are presumably esterified to the polymeric matrix. p-Coum-aric acid and lesser amounts of ferulic acid are found (Kolattukudy and Espelie, 1985). However, a recent reevaluation of the composition of suberin indicates that there is no solid evidence for aliphatic-aromatic linkages, nor is it understood how the phenolic units are joined, nor are their identies clear (Davin and Lewis, 1992). 

Mix equal volumes of bacterial cell supernatant and saturated solution MALDI matrix (67% water, 33% acetonitrile, 0.1-0.2% TFA) in an Eppendorf tube. Typical MALDI matrices for this application such as a-cyano-4-hydroxy-cinnamic acid (HCCA),sinapinic acid, and ferulic acid are mixed in an Eppendorf tube. 

The use of a highly acidified solution of ferulic acid as a MALDI matrix resulted in substantial modulation of the discriminatory power of MALDI-TOF MS profiling, which enabled successful discrimination of closely related species within the ACB complex. Microwave-assisted tryptic digestion represents a simple methodological variant that is accessible for routine MALDI-TOF MS profihng users. As the analysis was carried out using CHCA as a matrix, automatic mass spectral acquisition, which was not applicable for the previous alternative protocol employing acidified ferulic acid, was possible in this case.

The use of multicomponent matrices with accelerated sample drying along with a modified instrumental set-up was also considered when preparing samples for quantitative MALDI analysis. This method has been shown to improve the measured relative standard deviation (RSD) to better than 10% of analyte to internal standard (IS) intensity ratio [18]. Both, signal reproducibility and precision of the standard curve slope were improved by a factor of two when a DHB/fucose/5-methoxysaHcyUc acid multicomponent matrix was used compared to DHB alone. Similar results were observed when a feruhc acid/fucose multicomponent matrix was used compared to the use of ferulic add alone. 

Our laboratory used a number of different MALDI matrices (e.g., HCCA, sinapinic acid, ferulic add) for this work. HCCA is considered a hot matrix and favors the lower mass range generating singly, doubly.
