Fourth, the demonstration that many of these enzymes have prognostic value in cancer may qualify them as novel therapeutic targets. For example, similar to many other proteases, these enzymes may participate in the digestion of extracellular matrix, thus facilitating tumor invasion and metastasis. The identification of highly specific inhibitors may reveal new therapeutic opportunities. 

Heavily under-digested samples may be identified by inspection of the sample in the relevant double filter (e.g. Texas Red/FITC), see Figure 2.A. Under-digested samples are often characterized by apparent green autofluorescence in the cytosol as well as the extracellular matrix (Figure 2.A and C). 

FISH stains with under-digestion of tissue can be recognized due to autofluorescence in the cytosol and extracellular matrix and potential lack or reduced presence and intensity of red signals from the gene of interest (single locus). 

Numerous in vitro and in vivo tests have been developed to study specific steps of the neoangiogenic process. Several of these experimental protocols use the same techniques applied for assessment of tumor cell behavior (Albini, 1998). The activated endothelial cell acts similarly to a metastatic cell in the initial phases of angiogenesis, degrading the capillary basement membrane (BM), extravasating, digesting the extracellular matrix and moving toward the angiogenic stimulus (Liotta et al., 1991). Mammalian oocytes are surrounded by several layers of cells embedded in an extracellular matrix. This matrix has been shown to contain protein and hyaluronan [40].
