Aliquots (0.45 ml) of the euglobulin solution are transferred to test tubes, and 0.05 ml thrombin (Test Thrombin, Behring Werke) (25 U/ml) are added. The tubes are transferred to a water bath at 37 °C. The time interval between the addition of thrombin and the complete lysis of the clots is measured. 

Isoelectric precipitation and acid precipitation are also used to separate antibodies. Isoelectric precipitation (also called euglobulin precipitation) uses the solubility properties of a protein near its isoelectric point.69 When a concentrated protein solution in a low ionic strength buffer is titrated to its isoelectric point, it precipitates very slowly. Table 4 shows a balance of a process for purification of murine monoclonal IgM antibodies.70 The success of precipitation can be followed by the cumulative IgM content in the centrifugate and the sediment. The procedure is very gentle, but very sensitive to environmental conditions.

There are many synthetic chemicals that have been reported to increase lysis activity, chloroform being one of the earliest reported.68 More recently acid anti-inflammatory,87 antidiabetogenic,BS vasoactive,81 and diuretic agents,86 and anabolic steroids60 have been reported to have some effect on the lysis system. Even whiskey (alcohol) has been reported to enhance lysis.61 Hedner, et al.62 reported that ethyloestrend (8 mg/ day) increased spontaneous lysis in 45 patients with deep venous thrombosis and pulmonary embolism after 3 months of treatment. Euphillin (amino-phylline) was reported to increase abnormally low fibrinolysis in human patients and in rabbits with venous thrombosis.09 When 40 mg of furose-mide was injected i.v. into 33 healthy people, the euglobulin lysis time was shortened.64 Since this activation was not found in patients with uremia and nephrectomy, an intact renal system appears necessary.

Place the tubes in a water bath at 37°C and allow to stand for 2 min to attain temperature equilibrium. Using an automatic pipette, introduce into the bottom of the first tube 0.5 mL of a 1.0% m/V solution of bovine euglobulins R, ensuring mixing. At intervals of 5 s, introduce successively into the remaining tubes... 

Into a test tube 8 mm in diameter placed in a water bath at 37°C introduce 0.2 mL of a solution of a reference preparation of urokinase containing 1001.U. per milliliter and 0.1 mL of a solution of thrombin R containing 20 I.U. per milliliter. Add rapidly 0.5 mL of a solution containing 10 mg of bovine euglobulins per milliliter. A firm clot will form in less than 10 s. Note the time that elapses between the addition of the solution of bovine euglobulins and the lysis of the clot. The lysis time will not exceed 15 min. 

It is appropriate to consider the toxicities of different ions toward membranes. During freezing of thylakoids, anion toxicity decreases in the order 1 > Br > N03 > CT > F > acetate. This is reminiscent of the Hofmeister lyotropic power series, which was originally observed with regard to denaturation of euglobulins, then for blood clotting, then for...
