Ethylmalonic acid

Ethylmalonic Acid.—Like acetoacetic ester (see p. 83), diethylmalonate contains the gioup CO.CHj.CO. By the action of sodium or sodium alroholate, the hydrogen atoms of the methylene group are successively replaceable by sodium. The sodium atoms are in turn replaceable by alkyl or acyl groups. Thus, in the present preparation, ethyl malonic ester is obtained by the action of ethyl iodide on the monosodium compound. If this substance be treated with a second molecule of sodium alcoholate and a second molecule of alkyl iodide, a second radical would be in roduced, and a compound formed of the general formula... 

Butyric Acid from Ethylmalonic Acid.—A small distilling flask having a long side tube is fixed in an oblique position with the tube directed upwards. The ethylmalonic acid is placed in the flask, which is then corked and heated in an oil bath at 180° until, after half an hour, evolution of carbon dioxide has ceased. The residue is then distilled from the same flask in the usual way the butyric acid passes over between 162° and 163°. Yield 80-90 per cent of the theoretical.

When a reference standard is available and is calibrated against an internal standard in the TIC mode, six-point calibration curves of either individual or closely related analytes (for example, the isomers ethylmalonic acid, 2-methyIsuccinic, and glutaric acid) are run according to the standard/internal standard amount ratio shown in Table 3.1.5. Variations of this basic scheme may be indicated depending on the intensity of target ions ([M-15]+ species), the level of excretion in normal and disease states, and the linearity limit.

The ethylmalonic acid can also be worked up by precipitating its calcium salt from the neutralised solution by addition of a concentrated solution of calcium chloride, and treating the solid salt with cone, hydrochloric acid, extracting the liberated acid with ether, and proceeding as above. 

Ethylmalonic Acid.—The Irehavior of ethyl potassium ethylmalonate has already been mentioned in the discussion of malonie acid. The potassium salt, in t 20fslightly acid solution, yields propylene (Petersen ), and probably, like pyro tartarie acid, primary and secondary propyl alcohol. 

Hydroxymercuri methyl malonic methyl ester, IIOlIg.CMe (C02Me)2.—The preparation is carried out in a similar manner to tlie above, using equal molecules of the ester and mercuric oxide and continuing the shaking for several days. It is a white amorphous solid, insoluble in the usual solvents. When saponified as above it gives an 85 per cent, yield of the anhydride of h droxymercuri-propionic acid. In a similar manner the methyl ester of ethylmalonic acid yields 80 per cent, of hydroxymercuribiityric acid anhydride.

Ethylmalonic Acid. Stalder (S35) found ethylmalonic acid in normal urine of rat and man and showed that its excretion by the rat increases when isoleucine is fed. The increase is analogous to that of methylmalonic acid when valine is fed.
