Tolbutamide, drugs

Sulfonamides may inhibit the (hepatic) metabolism of the oral hypoglycemic drugs tolbutamide (Orinase) and chloq ropamide (Diabinese). This would increase the possibility of a hypoglycemic reaction. 

Among other antidiabetic drugs, tolbutamide was determined in formulations and in biological fluids by acid hydrolysis, reaction of the resulting amine with acetylacetone and formaldehyde, and fluorimetric determination of the resulting substituted dihydropyridine at 600 nm, with excitation at 480 nm (8).

The oxidation of the methyl group continues usually up to the carboxyl step (Fig. 19.11). This is observed for camphor, for 2-methyl-pyridine and for the drugs tolbutamide and alpidem, explaining the relatively short half-life of these latter compounds.
