Drug testing treatment

Erythromycin is considered the optimal drug for treatment of Campylobacter infections. The rate of resistance of Campylobacter to erythromycin remains low. Other advantages of this drug include ease of administration, low cost, lack of major toxicity, and narrow spectrum of activity.14 The recommended dosage for adults is 250 mg orally four times daily or 500 mg orally twice daily for 5 to 7 days. For very ill patients, treatment with gentamicin, imipenem, cefotaxime, or chloramphenicol is indicated, but susceptibility tests should be performed.

Placebo-controlled trial of a new active medicinal product, if positive, means that the trial was capable of detecting a difference, and that the test treatment is, at least, more efficacious than placebo. This achieves two outcomes provision of an internal validity check of the trial methods, and provision to regulatory authorities of a basis on which to judge the difference between a statistically significant but clinically inadequate effect that would probably lead to the drug not being licensed. 

Several tests with silicon containing compounds as therapeutics in human medicine have already been crowned with success. In France certain organosilicon preparations, DNR and RDN (compare Chap. 5.5), are used in the treatment of cardiovascular diseases, cancer and virus infections. In the Soviet Union extensive clinical tests with ointments of l-(chloromethyl)- and 1-ethoxysilatrane as possible drugs for treatment of different typs of alopecia were successful (compare Chap. 5.1). Further clinical studies showed that l-(chloromethyl)silatrane is also very promising in treatment of wounds and bums. In a Swedish hospital patients with poorly differentiated prostatic carcinoma were treated with 2,6-c/s-diphenyl-hexamethyl-cyclo-tetrasiloxane (Cisobitan ) the clinical study also yielded promising results (compare Chap. 5.3). 

Extensive clinical tests of l-(chloromethyl)silatrane and 1-ethoxysilatrane as possible drugs for treatment of alopecia have been carried out in several hospitals in Moscow, Kazan and Irkutsk71-74). The silatranes were used as 3-5% ointments, creams, and liniments on different, specially chosen bases. 

At three-months, the stenosis was reduced by 20 and 34% in the treatment groups. Histopathology evaluation showed that there were no adverse effects of the drug-loaded stent compared to the controls, and no deleterious phenomenon could be attributed to the drug tested.
