The measurement of contact angles for a sessile drop or bubble resting on or against a plane solid surface can be measured by direct microscopic examination. 

The most important property characterizing a microporous membrane is the pore diameter (d). Some of the methods of measuring pore diameters are described in Chapter 7. Although microporous membranes are usually characterized by a single pore diameter value, most membranes actually contain a range of pore sizes. In ultrafiltration, the pore diameter quoted is usually an average value, but to confuse the issue, the pore diameter in microfiltration is usually defined in terms of the largest particle able to penetrate the membrane. This nominal pore diameter can be 5 to 10 times smaller than the apparent pore diameter based on direct microscopic examination of the membrane. 

There are two methods of estimating the number of cells in a suspension. Using a haemocytometer the number of cells in a given volume is counted by direct microscopic examination. Using electronic counters, e.g. the Coulter counter, the cells in a given volume of suspension are drawn through an orifice and registered electronically. 

Identification of many particles and aerosols can be made using direct microscopic examination coupled with a knowledge of the collection circumstances. Recovery of C. immitis from infected tissues or secretions for direct examination and culture provides an accurate and rapid method of diagnosis. Direct microscopic examination and histopathologic studies of infected tissues will reveal the large, mature endosporulating spherules. Young spherules without endospores may be confused, however, with other fungi. Silver stains of body fluids or tissue biopsies are also helpful. 

Various methods for counting bacteria in milk by microscopic examination have been described, but the method that is commonly described as a direct microscopic examination of a dried film of milk has been found to be the simplest and most reliable method of counting the bacteria as they exist in the milk itself. It is recognized in this report as a standard or official technique of equal standing with the colony count from agar plates where used for judging the quality of unpasteurized milk. 

It must swell extensively in the solvents used for synthesis, allowing all reagents to penetrate readily throughout the particles of the polymer.
