Dendritic cells enter lymph nodes through their CD49d-pi integrins at high-endothelial venules. There are two populations of dendritic cells CDllc andCDllc". Although speculative, these cell surface markers may be responsible for targeting dendritic cells to T-lymphocyte areas or B-lymphocyte areas (67).

The lymph node harbors a composite population of dendritic cells (DCs)— residents and emigrants—capable of selection, stimulation and elimination of effector lymphocytes. These cells trigger adaptive immunity and/or tolerance. In the human lymph node T-cell compartment, two primary subsets of DCs, compared to six in mice, have been identified monocytoid CDl lb CDl Ic" and plasmacytoid interferon (IFN)-a producing CD123" CDllb CDllc". The last subset clusters with T cells around HEVs.

In humans, CDllc -dendritic cells promote Thl responses, whereas dendritic cells derived from CDllc" precursors induce Th2 responses. The degree to which Th polarization occurs may differ according to the maturation state of the dendritic cell and the ratio of dendritic cells to T-lymphocytes (66,67).
