Figure 26-21 shows a typical student-grade benchtop FTIR spectrometer that has a built-in computer for data acquisition, analysis, and presentation. The instrument i.s relatively inexpensive (approximately 11,000), has a resolution of 4 cm. ... [Pg.813]

The initial instrument qualification process is detailed and comprehensive and includes both software (computer programs used in data acquisition, analysis, and archiving) and hardware (pumps, detectors, injectors, etc.). Once the instrument is qualified then methods themselves are developed and validated. The development process includes the initial evaluation of the appropriateness of the test, which is often determined by the ability of the method to meet targeted method requirements or mandates (linear range, linearity, accuracy, quantitation limits, resolution, etc.). Once these fundamental requirements are met, the cost effectiveness and overall efficiency are reviewed. Upon acceptance, a developed method then goes through a validation process. [Pg.63]

Samples were run on a Beckman P/ACE System 5010 CE unit (Beckman Instruments, Palo. Alto. Calif.). A 57 cm x 75 pm i.d. (Beckman Instruments, Palo. Alto. Calif), a 57 cm X 50 pm i.d. capillary and a 47 cm x 50 pm capillary (Polymicro Technologies) were used. Data acquisition, analysis, and instrument control were done with Beckman System Gold software. Unless otherwise noted, all samples were run at 23 C and were injected hydrodynamically for 5 seconds with 0.5 psi pressure. This corresponds to a 30 nL injection for the 57 cm x 75 pm i.d. capillary, 8.0 nL for the 47 cm X 50 pm capillary and 6.0 nL for the 57 cm x 50 pm i.d. capillary. The voltage and run time varied depending on the sample, capillary and are noted in the text. UV detection was performed on-column (deuterium lamp, 200 nm).
