Tissue factor pathway inhibitor (TFPI) is a major physiologic inhibitor of coagulation. It is a protein that circulates in the blood associated with lipoproteins. TFPI directly inhibits factor Xa by binding to the enzyme near its active site. This factor Xa-TFPI complex then inhibits the factor Vlla-tissue factor complex. [Pg.601]

Hemostasis and thrombosis are complex processes involving coagulation factors, platelets, and blood vessels. [Pg.608]

Once injured or activated by a toxic substance (e.g., bacterial toxins, placenta chemicals, snake venom, etc.), endothelial cells and monocytes respond by generating tissue factor on the cell surface. This, in turn, leads to the generation of tissue factor-factor Villa complexes, followed by unregulated and excessive generation of thrombin, fibrin, systemic microthrombi and consumption of coagulation factors,... [Pg.996]

Complex coagulation derangements can occur in cirrhosis. These derangements include the reduction in the synthesis of coagulation factors, excessive fibrinolysis, disseminated intravascular coagulation, thrombocytopenia, and platelet dysfunction. [Pg.253]

Factor VIII (FVIII) is an essential coagulation factor in the blood which serves as a cofactor in the complex blood-clotting cascade. A deficiency in FVIII is the... [Pg.669]

Considerably more is known about anticoagulants from the blood-feeding hookworm, Ancylostoma caninum. Several families of small, anti-clotting peptides have been described in A. caninum. AcAP, an 8.7 kDa peptide secreted by adult worms that does not exhibit homology to other anticoagulants, but is a potent inhibitor of Factor Xa, was described by Cappello et at. (1995). A family of related peptides, one of which uniquely inhibited a complex of blood coagulation factor VIIA and tissue factor, was also reported (Stanssens et al., 1996). [Pg.352]

Schematic view of the role of coagulation factor Xa in arterial thrombosis. After endothelial injury, platelets adhere to the subendothelial matrix. The procoagulant activity of the arterial clot can be attributed to the formation of the prothrombinase complex on the platelet surface which cleaves prothrombin and produces thrombin. Thrombin subsequently acts as a strong agonist of further platelet aggregation. Schematic view of the role of coagulation factor Xa in arterial thrombosis.
