In our laboratory, the viability of excised porcine buccal mucosa was assessed using histological evaluation and a 3-[4,5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) biochemical assay which has previously been used in assessing the viability of excised buccal mucosa and cornea [49, 50], Histological evaluation of tissue demonstrated that the buccal epithelium appeared viable up to 9 h postmortem, and this was supported by the MTT biochemical assay, which indicated that viability was maintained for up to 12 h [80], Therefore, we recommend that all permeation experiments using porcine buccal mucosa be performed within 9-12 h of animal death.

Human oral mucosa consists of different cell types including keratinized and non-keratinized striated epithelial, but the buccal mucosa is composed predominately of the latter. In selecting an appropriate animal model care was taken to ensure that the mucosal structure in the selected species matched that in man as closely as possible. Based on histological examinations all the rodent species (rat, guinea pig, hamster and rabbit) would constitute poor models because of extensive keratinization of their buccal mucosa. Of the other possibilities, the dog appeared to be the best choice. 

Basaloid squamous cell carcinoma (BSCC) is an uncommon, histologically distinct variant of SCC. In the upper aerodigestive tract, it occurs frequently in the base of the tongue, tonsils, hypopharynx, and larynx but has also been described in many other locations such as the palate, buccal mucosa, floor of mouth, nasopharynx, nasal cavity, and trachea. This is one of the subtypes of squamous carcinoma that can present with large neck node metastases and an unknown primary tumor.
