Baseline determinations

As a consequence of the development of extraction methods for STA based on mixed-mode SPE columns, as well as of the recent introduction of instruments for the automated sample preparation allowing efficient evaporation and derivatization of the extracts, full automation of STA methods based on GC-MS analysis is also available. It needs GC-MS instalments equipped with an HP PrepStation System. The samples directly injected by the PrepStation are analyzed by full scan GC-MS. Using macrocommands, peak identification and reporting of the results are also automated. Each ion of interest is automatically selected, retention time is calculated, and the peak area is determined. All data are checked for interference, peak selection, and baseline determination.

Example 2.10 Suppose the researcher exposed agar plates inoculated with Escherichia coli to forearms of human subjects that are treated with an antimicrobial formulation, as in an agar-patch test. In the study, four plates were attached to each of the treated forearms of each subject. The agar plates were removed from 24 h of incubation at 35°C 2°C, and the colonies were counted. The duplicate plates at each time point for each subject were averaged to provide one value for each subject at each time. 

Baseline determination is one of the most difficult emd important tasks in peak-area allocation, since the location of the baseline greatly influences the value of the areas. Except for some simple algorithms, there are few details in the literature about baseline construction. 

The initial baseline determination partially considers the nature, extent, circumstances, and gravity of the alleged violation. 

In addition to the study of electrochemical processes at representative electrodes, in situ FTIR spectroscopy has been slow in evolving as a routine, quantitative analytical tool, primarily as a result of the difficulties associated with baseline determination.

There is a variable that accounts for the baseline stability. Increasing this variable allows for a number of sudden but small changes in the response to have less effect on the baseline determination.
