Antischistosomal drugs

Several other heterocyclic compounds are used as veterinary anthelmintics although they lack the spectrum of activity of the above classes. For example, the antischistosomal drug, praziquantel (14), is highly effective in the treatment of cestodes in animals and man. The piperazine-derived drug diethylcarbamazine (15a) is used as a filaricide and for treatment of lungworm infections in sheep and cattle. The mode of action of diethylcarbamazine (13a) is uncertain but recent studies suggest that an effect on the helminth cuticle may be important. [Pg.203]

The 5-nitrofuryl group225 is promulgated as an efficacious moiety to ensure chemotherapeutic value, and the trans derivative (135) was found to be active against Staphylococcus aureus,171 It is also a broad-spectrum antimicrobial agent226 and an antischistosomal drug.227 The latter activity is due to reduction of the phosphorylase phosphatase in the worms themselves, damaging reproduction in the female.227, 228... [Pg.115]

S-transferases are implicated in resistance to certain antiparasitic drugs. One form of S. mansoni GSH S-transferase (SmGST-5) catalyzes the GSH-dependent demethylation of dichlorvos, the active form of the antischistosomal drug metrifonate (50). The two... [Pg.168]

Antimony-based compounds have been used to treat certain protozoan parasitic diseases. The potassium tartrate and sodium tartrate forms of antimony have been used extensively in the past on a global basis as antischistosomal drugs (Cooper and Harrison 2009 ATSDR 1992), and the pentavalent form of antimony, Sb(V), also has been used to treat leishmaniasis (Cooper and Harrison 2009). Leishmaniasis is caused by protozoan parasites of the Trypanosomatidae family. It has been estimated that leishmaniasis affects 2 million people on a global basis with a mortality rate in excess of 70,000 persons per year (Baiocco et al. 2009). Mechanistically, trivalent antimony, Sb(lll), is known to interfere with trypanothion reductase, an enzyme that plays an important role in the survival of the trypano-somatid parasite. [Pg.207]
