Antipruritic agents

The compounds of the invention were evaluated as antipruritic agents by measuring their ability to inhibit hind leg scratching induced in rats using a known pruritogenic agent according to the method of Berendsen (1). 

In a separate investigation by the author (4) Step 2 benzoate derivatives, (II), were prepared and were also effective as opioid receptor antagonists and used as antipruritic agents. 

The treatment for these disorders is limited. Antipruritic agents and UDCA are indicated for itching. Good dietary management lowers the risk of malnutrition. Bi-phosphonates and calcium can have a positive effect on bone density. As yet, there is no medical treatment available for the often annoying chronic fatigue syndrome. Liver transplantation may ultimately be indicated. 

Cyproheptadine po.sscs.ses both antihistamine and antisc-rotonin activity and is used as an antipruritic agent. Sedation is the most prominent side effect, and this is usually brief, disappearing after 3 or 4 days of tieatmcnt.
