Electrophysiological studies in the hippocampus also support enhanced al responses after chronic antidepressant treatments. Recent data indicate that the novel antidepressant tianeptine, which may increase serotonin reuptake when given chronically, also increases responsiveness of the al-adrenergic system (Rogoz et al. 2001). [Pg.213]

In contrast to SSRIs, the atypical antidepressant tianeptine has been reported to enhance uptake of serotonin and has been proposed to act by restoring neuroplasticity (Costa e Silva, 2004). A double-blind crossover trial of tianeptine vs. placebo in 12 autistic children reported modest but statistically significant improvements after 6 weeks of treatment with respect to ABC measures of irritability, hyperactivity, inadequate eye contact, and inappropriate speech. The major side effects noted were drowsiness and decreased activity (Niederhofer et al., 2003). Larger studies of this agent in ASD populations would be useful. [Pg.249]

Considerable attention has been focussed on the antidepressant, tianeptine (23), which has included the development of a reverse-phase, ion-pair, liquid chromatographic method for the determination of the drug and its major metabolites in plasma, urine, and tissues <86JC115>. Further metabolic studies in healthy volunteers involving chromatographic and mass spectral analysis have also been reported <90mi 907-01 >. [Pg.186]

The behavioural effects of two antidepressants with opposite molecular actions, ie. tianeptine (a serotonin reuptake enhancer) and fluoxetine (a serotonin reuptake blocker) have been assessed and it was concluded that, apart from the effects on serotonin reuptake, these dmgs have other mechanisms playing an important role in the anti-depressant action <00AF5>. [Pg.373]

Conformational aspects of the antidepressant drug, tianeptine (23), which is a dibenzothiazepine derivative, have been studied by H and C NMR spectroscopy. Two separate conformations for the seven-membered ring are indicated at low to room temperature. The conformational preference for the side chain involves bending over the non-substituted aromatic ring moiety <92MRC1212>. [Pg.186]

Tianeptine, (K5)-7-(3-chloro-6-methyl-6,ll-dihydrodibenzo[c/ [l,2]lhiazepin-ll-yl-amino)heptanoic acid 5, 5-dioxide, is an antidepressant effective against anxiety accompanying mood disorders. The drug is used primarily in the treatment of major depressive disorder, although it may also be used to treat asthma or irritable bowel syndrome. The TLC method was used to separate the photodegradation products of tianeptine [16] (Figure 14.13). [Pg.251]
