Antibacterial agents systemic synthetic

The antibacterial agent flumequine 280 was synthetized in optically active form by starting with resolution of the two enantiomers of a suitably substituted racemic tetrahydroquinoline through formation of the (lf )-3-bromocamphor-8-sulfonates. After N-alkylation of the (2K)-tetrahydroisoquinoline enantiomer 277 with diethyl ethoxymethylene-malonate to give 278, the quinolizidine system 279 was formed by acylation onto the peri-position. This compound was finally hydrolyzed to afford 280 (Scheme 60) <1999TA1079>. [Pg.41]

Since the introduction in 1963 of nalidixic acid (Table I), l-ethyl-1,4-dihydro-7-methyl-4-oxo-l,8-naphthyridine-3-carboxylic acid, as a systemic Gram-negative antibacterial agent, a large number of analogues have been synthesized and evaluated, some of which have come onto the international market. CL2) A comprehensive review has outlined the synthetic methods, microbiology and structure activity relationships of those derivatives reported prior to 1977.(2)... [Pg.301]

Chloramphenicol. Chloramphenicol (Chloromycetin) is a synthetically produced agent that exerts antibacterial effects similar to those of erythromycin that is, it binds to the 50S subunit of bacterial ribosomes and inhibits peptide bond formation. Chloramphenicol is a broad-spectrum antibiotic that is active against many gram-negative and gram-positive bacteria. This drug is administered systemically to treat serious infections such as typhoid fever, Haemophilus infections such as osteomyelitis, rickettsial infections such as Rocky Mountain spotted fever, and certain forms of meningitis. Chloramphenicol may also be administered topically to treat various skin, eye, and ear infections. [Pg.509]

Derivatives of all three heterocyclic systems have been widely investigated for use in synthetic drugs (see also above) amongst the most commonly used compounds are the antibacterial Trimethoprim, the antimalarial Pyrimethamine and the anti-hypertensive agent Hydralazine (containing a phthalazine nucleus). Piperazine (hexahydropyrazine) is used in the treatment of intestinal nematode (worm) infections. [Pg.190]

Dendrimers, dendrons and other dendritic systems have been synthesized as interesting compounds with a great potential in different biomedical applications, including delivery carriers of drugs, development of synthetic vaccines, antitumor systems or as therapeutic agents per se with, for example, antiviral and antibacterial properties. [Pg.347]

Antiviral Antibiotics - Screening fermentation mixtures, identifying known compounds, and assaying samples during the purification of active fermentation products, are considered to be more difficult than the examination of pure synthetic compounds as antiviral agents. However, a review of the development of useful antibacterials would indicate that the increased effort is Justified. While most antibiotics isolated as antibacterial substances will eventually be tested in antiviral systems, it seems valid to expect certain cultures to produce specific antivirals which are not inhibitors of other microorganisms. In a series of papers on antiviral and antitumor antibiotics, Takatsuki have pre-... [Pg.106]
