Anilines, diphenylamine

In the derivatization of sugars with aniline-diphenylamine reagent for example, this leads to unsatisfactory irregular coloration. The standard deviation for the method deteriorates from 2 to 3% to 5 to 8%. For this reason color reactions should be avoided for direct quantitation if it is possible to scan in the UV range without derivatization. [Pg.96]

To detect glycosides heat the chromatograms to 130—150°C for 15 min. Blue-grey zones are produced (detection limit prunasin 0.3—0.5 pg [18]). Flavonoids are better detected with a modified reagent of the following composition phosphoric acid (85%) — acetic acid — aniline — diphenylamine (20 ml - -100 ml + 5 ml + 5 g). [Pg.181]

TLC and HPLC. To determine the best solvents for HPLC, 5pl of the C18 concentrated PVP fraction were separated on analytical TLC plates coated with either C18 reversed phase adsorbant or with silica. One cm sections of the plates were removed, placed in buffer, hydrolyzed, extracted and the p-damascenone charm determined. Standards containing 5 mg each of 1 -O-n-octyl- p-D-glucopyranoside and a-methyl-D-mannoside were run simultaneously on the same plate and visualized by spraying with aniline-diphenylamine-acetone-phosphoric acid 80% (4 mL 4 g 200 mL 30 mL) (10). HPLC separations were performed on a 0.46 mm x 25 cm column packed with 5 im C18, eluted with 60/40 methanol water at a flow of 0.8 mL/min. The refractive index of samples was recorded with a Knauer differential refractometer. Fractions were collected every minute and assayed for p-damascenone charm. [Pg.80]

A system for the screening and identification of reducing sugars in urine using one-dimensional t.l.c. on silica gel has been developed.Colour was developed by the aniline-diphenylamine method. [Pg.239]

Thin-layer plates may be developed by spraying with aniline-diphenylamine-acetone-80% phosphoric acid (4 ml 4 g 200 ml 30 ml) followed by heating at 105°C for 30 min. The combination of the isotope technique with PC is still one of the best methods, not only for the detection of sugar alcohols but also for the determination of their turnover and elucidation of their biosynthesis. Radioactive spots are eluted from chromatograms, evaporated on to plan-chettes, and the radioactivity measured with a counter. [Pg.455]
