Methylamination of 2-amino-3-nitropyridine gives in good yield 2-amino-6-methylamino-3-nitropyridine, while methylamination of 6-amino-3-nitro-pyridine only gives (in a small yield) 6-amino-2-methylamino-3-nitropyr-idine (Scheme 7). The activating influence of the 3-nitro group on the C-6 para position explains this difference in reactivity.

As discussed before, in liquid ammonia/potassium permanganate nucleo-phugal substituents at C-2, such as ones present in the naphthyridines (84c, 84e, 84h, 84i, 841, and 84m), could not be replaced by the amino group only SnH substitution takes place. However, it has been observed that in the reaction of the 2-chloro-3-nitro-l,8-naphthyridine (92c) with liquid methylamine/potassium permanganate S H substitution as well as methylamino-dechlorination takes place, yielding 2,4-bis-(methylamino)-3-nitro-l,8-naphthyridine (93c). 

It has been prepared synthetically by Ewins in the following manner Meta-oxybenzoic acid is converted with the aid of dimethyl sulphate into m-methoxybenzoic acid, which is then nitrated, and from the nitration products 2-nitro-3-methoxybenzoic acid is separated. This is reduced to 2-amino-3-methoxybenzoic acid which on heating with methyl iodide, yields 2-methylamino-3-methoxybenzoic acid. On warming this with freshly precipitated silver chloride it yields damascenine hydrochloride.
