Although the isolation of the principal alkaloid, ibogaine, of Taber-nanthe iboga was described at the turn of the present century (1), it was not until the early 1950 s that serious work on its structure was begun. It had been shown to contain a methoxy group and, by means of color reactions (19) and by measurement of its UV-spectrum, to be an indole (20), but it was not recognized to be a methoxyindole until permanganate oxidation was found to afford 5-methoxy-A-oxalylanthranilic acid (21). [Pg.206]

Another example is the indole alkaloid ibogaine. Radioligand binding assays revealed that ibogaine interacted with a wide variety of receptors at concns. of 1-100 p.M. These included the opiate, 5-HT2, 5HT3, and muscarinic [, 2 receptors, and the dopamine, norepinephrine, and serotonin uptake sites. Furthermore, ibogaine interacted with N-methyl-D-aspartic acid (NMDA) associated ion and sodium ion channels. The alkaloid ibogaine (LIV), despite being a formal derivative of skeletal pattern XXII whereas yohimbine is a formal derivative of skeletal pattern XXI, shows similar activity to yohimbine in so far as it produces anxiety in normal individuals and induces a psychotic state in schizophrenics [1061, 1078, 1080-1082] as well as raising the blood pressure in conscious animals [1061, 1083] but not in anaesthetized animals [1061, 1083, 1084].
