Adaptation to culture

The NADH dehydrogenase of yeast is of considerable interest because in Saccharomyces cerevisiae and Saccharomyces carlsbergensis coupling site 1 is absent, whereas in Candida utilis its existence depends on the growth phase of the cells and can be altered by adaptations to culture conditions and by catabolite repression. 

To attain consistent and stable production of FVIII from CHO cells adapted to culture in a protein-free medium, continuous (chemostat) perfusion is used in the bioreactor culture system. An important advantage of the system is that the culture conditions can be continuously controlled, monitored, and optimized. 

Once ten to fifteen fruiting strains are In hand, they should be tested In a small scale version of the ultimate culture method. This step allows the strain best adapted to culture conditions to be selected. All strains should be tested at least In duplicates five replicates per strain are preferable.

Andersen et al. (1996) and Andersen (1995) have studied the effect of temperature on the recombinant protein production using a baulovinis/insect cell expression system. In Tables 17.15, 17.16, 17.17, 17.18 and 17.19 we reproduce the growth data obtained in spinner flasks (batch cultures) using Bombyx mori (Bm5) cells adapted to serum-free media (Ex-Cell 400). The working volume was 125 ml and samples were taken twice daily. The cultures were carried out at six different incubation temperatures (22, 26,28, 30 and 32 TT).

The biochemical population can be specifically adapted to particular pollutants. However, in the majority of cases, a wide range of organic materials must be dealt with and mixed cultures are used. 

HeLa S3 (ATCC CCL-2.2) cells, a clonal derivative of the parent HeLa line (ATCC CCL-2), which are adapted to grow in suspension and therefore more suitable for large biomass production, are used for the preparation of human cell extract. The cells are maintained in suspension culture in Coming 850 cm2 Polystyrene Roller Bottles at 37° at a concentration of 3 to 6 x 105 cells/ml in Eagle s Minimum Essential Medium Joklik Modification (Sigma) supplemented with 10% Fetal Bovine Serum (Invitrogen) in the presence of 5% C02.
