Acetylsalicylic acid synthetic derivatives

Little information is available regarding tissue distribution or metabolic products of nonsteroidal anti-inflammatory drugs in cattle. The early synthetic compounds were simple derivatives either of salicylic acid such as acetylsalicylic acid and methylsalicylic acid, or of pyrazolone such as metamizole, oxyphenbuta-zone, phenylbutazone, propylphenazone, and suxibuzone. Sufentanil is a synthetic opioid analgesic drug, as outlined in CHEC-II(1996), and is approximately 5-10 times more potent than fentanyl. Sufentanil is marketed for use by specialist centers under different trade names. Other thiophene derivatives, such as the benzimidazol-thiophenes 16 and 17, exhibit analgesic activity that can be compared with morphine sulfate and acetylsalicylic acid <2005PS1841>.

Other routes for the preparation of such diones, such as the pyrolysis of acetylsalicylic acid oligomers used for the preparation of disalicylide itself <82Mi 924-02) and the dimerization of salicylic acid derivatives <88CZ125, 9iJOC4525>, presumably would at best yield mixtures if applied to unsymmetrical analogues.

A) Acetylsalicylic Add (Aspirin). Salicylic acid and its derivatives were among the first synthetic organic compounds to be used for therapeutic purposes. The sodium salt of salicylic acid, the methyl and phenyl esters, and the acetyl derivative of salicylic acid are extensively used to diminish pain (as analgesics) and to reduce the temperature of the body (as antipyretics). The most widely used is the acetyl derivative which is sold under the name aspirin. Salicylic acid is prepared industrially by the carbonation of sodium phenoxide, at 180-200° at atmospheric pressure or 120-130° at a pressure of a few atmospheres.
