The mature seed of this plant contained 6-10% of 5-hydroxytrypto-phan. An enzyme system capable of hydroxylating tryptophan was identified in various tissues. 5-Hydroxytryptamine (up to 0.2%) was found in the pods and in lesser amounts in other tissues. 5-Hydroxy-indole-3-acetic acid and indole-3-acetylaspartic acid were also identified 122).

Application of [2- C]IAA to the cotyledon of etiolated 5-day-old broad been (Vicia faba L. cv Chukyo) seedlings resulted in accumulation of radioactive substances in the root primordia and in the stele of the basal part of the roots [24, 25]. Two metabolites being more polar than lAA-Asp accounted for 70-80% of total radioactivity in the root after 24-h treatment, and they were not extracted with ether in acid pH. After hydrolysis with 2 M HCl or 7 M NaOH, their radioactive moieties were extracted with ether, but they did not coincide with lAA. We purified the two substances from Vicia roots and identified them as 3-(0-)S-glucosyl)-2-indolone-3-acetylaspartic acid (Glc-DIA-Asp) and 3-hydroxy-2-indolone-3-acetylaspartic acid (DIA-Asp) [26, 27]. The DIA moiety is converted into 2-quinolone-4-carboxylic acid (QCA) by acid hydrolysis and the UV spectrum of QCA is quite different from that of DIA, which is in contrast with the conversion of OxIAA into l,2,3,4-tetrahydro-2-quinolone-4-carboxylic acid without accompanying large spectral change. 

Figure 1 shows the metabolic pathway of lAA in K/c/Vz seedlings. The precursor of the DIA conjugates was investigated using [2- C]DIA, indole-3-[2- C]acetylaspartic acid and indole-3-[2- C]acetyl[2,3- H]aspartic acid [28]. The radioactive substances were applied to the cotyledon or to the basal part of the roots of Vicia seedlings, and their metabolites were analyzed by TLC and reversed phase Ci8 HPLC. [ C]DIA was not metabolized, but [ C]IAA-Asp was converted into...

Canavan disease usually presents in early childhood with hypotonia, visual loss and accelerated head growth. MRI of the brain shows a leukodystrophy. In urine, plasma, and CSF N-acetylaspartic acid is elevated. However, sometimes the elevation in urine is marginal. The diagnosis can be confirmed by analysis of aspartoacylase and by mutation studies. Prenatal diagnosis is possible, preferably by the combined analysis of N-acetylaspartic acid in amniotic fluid and mutation studies.
