All these observations underscore the potential for application of appropriate OPAs to the destruction of organophosphorus compounds with anticholinesterase activity (Cheng and Calomiris 1996). However, since, hydrolysis results in release of fluoride, the possibility of its subsequent incorporation into organic substrates to produce fluoroacetate and 4-fluorothreonine (Reid et al. 1995) may be worth consideration. 

A principle of the therapy for acute poisonings with anticholinesterase compounds (OPC, carbamates) lies in the complex performance of specific antidotic therapy including methods for poison excretion and intensive resuscitation measures.

Satisfactory neuromuscular block can be maintained with an atracurium bolus of 0.5-0.6 mg-kg-1 followed by an infusion at a rate of about 0.5 mg-kg-l-h-1. The unique metabolism of atracurium makes it suitable for use in critically ill patients with organ dysfunction, in whom it can be used for even longer periods of time as long as hypothermia is not present. The reversal of an atracurium block can be easily accomplished with anticholinesterase agents. 

Treatment of test animals with anticholinesterase agents such as atropine and 2-PAM significantly reduced the acute lethality of diazinon in rats indicating that acute diazinon lethality is primarily attributable to the inhibition of acetylcholinesterase. Administration of 16 mg/kg atropine intramuscularly, with or without 30 mg/kg pyridine 2-aldoxime methochloride (2-PAM) given either orally or intravenously or both, to female albino rats 10 minutes before diazinon exposure increased the LD50 value (294 mg/kg) for diazinon for this species by a factor of 3.2 (with 2-PAM) or 1.7 (without 2-PAM) (Harris et al. 1969).

Subjects tested with anticholinesterases, anticholinergics, cholinesterase reactivators, or psychochemicals did not differ significantly from NGT or OCT subjects in their replies to questions about current health status. Almost 90% of all respondents reported no health problems related to toxic exposures, and 79% reported good to excellent health. (Subjects tested with LSD were not within the purview of this investigation, inasmuch as they had been evaluated and reported on earlier by a different group, which used other methods.

The diagnostic evaluation of myasthenia gravis includes a complete history and physical examination, objective evidence of circulating acetylcholine receptor antibodies, electrophysiologic evidence of abnormal neuromuscular transmission, and pharmacologic evaluation with anticholinesterase drugs. 

Reversal of this type of neuromuscular block can be achieved with anticholinesterase drugs, such as neostigmine, which prevent the destruction by cholinesterase of acetylcholine released at nerve endings, allow the concentration to build up and so reduce the competitive effect of a blocking agent.

Rick There is. I believe, a species-specific effect in hypothermia and hyperthermia. This has been shown by Feldberg and Myers (1964) and others in their work with catecholamines. To what extent is this species-specific effect on thermoregulation present with anticholinesterases ... 

Anti-inflammatory corticosteroids, for example prednisolone, can be used to suppress the antibody formation in myasthenia gravis. They should be used together with anticholinesterase drugs and once an improvement is seen the dose should be decreased.
