Wheat germ structure

More-detailed studies of enzyme specificity towards the structure of the nucleoside residue have been performed with the sucrose synthetase of pea seedlings,339,384,501,502 and with wheat-germ arbutin synthetase.339,364,503 Neither enzyme can use the cytidine,504 isocyti-dine, or N3-methyluridine derivatives as substrates. On the other hand, the a-D-glucopyranosyl pyrophosphate esters of 4-thiouri-... [Pg.394]

In a further experiment we assayed for the presence of a cap structure on the mRNAs for both Inhibitors I and II by competitive inhibition by 7-methyl-guanosine 5 -monophosphate (m G p) of the in vitro translation of these messengers. Concentrations of 40 pM m G p inhibited by 50% the in vitro translation of total tomato leaf poly(A)" " mRNA (Fig. 7A). This level is 40-fold lower than that required to similarly inhibit rabbit globin mRNA translated in a rabbit reticulocyte lysate (17) and 4-fold lower than that required to inhibit the same mRNA in a wheat germ system (18). It was of interest that the translation of Inhibitor I is inhibited to 50% by 20 pM m G p while 50% inhibition of Inhibitor II requires less than 10 pM (Fig. 7B). The basis of this difference is not understood but... [Pg.116]

The CFCF described by Spirin (30) is efficient for protein production using the efficient wheat germ cell-free system, as shown in the previous section. However, this system may not be suitable for massive screening of cDNA libraries for structural genomics and production of gene products. Because the CFCF apparatus is equipped with a semipermeable membrane in the reaction chamber, the system is quite complicated to manipulate. The delicate nature of the membrane can pose problems for automating the translation process. Also,... [Pg.162]

The simplest type of phosphoinositide is represented by the phosphatide isolated from horse liver or dog liver by McKibbin.171 This consists only of glycerol, myo-inositol, phosphoric acid, and fatty acids it probably has structure LXXX. Similar inositides, differing only in the nature of the fatty acids, have been isolated from wheat germ,187 beef heart, 188 and beef liver.189 That the major portion of the fatty acids is attached to glycerol, not to myo-inositol, was shown by the isolation of a diglyceride on mild hydrolysis.189... [Pg.175]
