Propoxyphene is chemically related to methadone but has low analgesic activity. Various studies have reported its potency at levels ranging from no better than placebo to half as potent as codeine that is, 120 mg propoxyphene = 60 mg codeine. Its true potency probably lies somewhere between these extremes, and its analgesic effect is additive to that of an optimal dose of aspirin or acetaminophen. However, its low efficacy makes it unsuitable, even in combination with aspirin, for severe pain. The increasing incidence of deaths associated with its use and misuse has caused it to be scheduled as a controlled substance. Moreover, banning its use in the United States is under consideration. [Pg.701]

Two statistical terms involving error analysis that are often used and misused are accuracy and precision. Precision refers to the extent of agreement among repeated measurements of an experimental value. Accuracy is defined as the difference between the experimental value and the true value for the quantity. Since the true value is seldom known, accuracy is better defined as the difference between the experimental value and the accepted true value. Several experimental measurements may be precise (that is, in close agreement with each other) without being accurate. [Pg.26]

A wide range of possible uses and misuses must be considered in both the development of formulations and in regulatory decisions. Formulations need to be tested as they might be used, not just as they are meant to be used. While it is possible to develop formulations that lower the abuse potential of a pharmacological constituent, every effort must be made to challenge the formulation to substantiate such a claim. [Pg.151]
