Urine Tamm-Horsfall glycoprotein

The normal urinary total protein excretion is less than 150mg/24hr. The proteins excreted are made up of mostly albumin (50% to 60%) and some smaller proteins, together with proteins secreted by the tubules, of which Tamm-Horsfall glycoprotein (THG) is one. The normal concentrations of proteins found in urine are listed in Table 24-1. 

Tamm-Horsfall glycoprotein has been isolated from calf urine. Mild acid hydrolysis cleaved the glycoprotein into two lower molecular weight fragments (mol. wts. 6.6 X 10 and 5.1 X 10 ). 

Cultures of baby-hamster kidney cells adapted to grow in suspension produced and released a glycoprotein that is similar to, if not identical with, the Tamm-Horsfall glycoprotein in hamster urine. The amount of this glycoprotein increases during the G2 phase of the cell-cycle and then decreases before physical division of the cells.

The salt-soluble fraction is the supernatant liquor obtained on treatment of urine with 0.58 M sodium chloride to precipitate the Tamm-Horsfall acid glycoprotein.22... 

A glycoprotein resembling the ai-glycoprotein of blood serum has been isolated from the urine of patients with proteinuria 85). It contains D-glu-cosamine, D-mannose, and D-galactose as its carbohydrate constituents. Its electrophoretic behavior and immunochemical reactions greatly resemble those of the ai-glycoprotein isolated by Schmid. It is clearly not the glycoprotein isolated from normal urine by Tamm and Horsfall.
