Ultraviolet spectroscopy chromatographic detection

Tor reference. Positive identification can be made only by collecting the compound or transierring it as it elutes directly into another apparatus for analysis by other means, such as infrared or ultraviolet spectroscopy, mass spectrometry, or nuclear magnetic resonance. Commercially available apparatus is available which combines in a single unit both a gas chromatograph and an infrared, ultraviolet, or mass spectrometer for routine separation and identilicalion. The ancillary system may also be microprocessor-based, with an extensive memory for storing libraries of known infrared spectra or fragmentation patterns (in the case of mass spectrometers). Such systems allow microprocessor-controlled comparison and identilicalion of detected compounds. [Pg.378]

Chromatographic techniques are the main methods that have been used to separate, detect, and identify organic components of FDR.152 162 Other methods considered include molecular luminescence,163 infrared spectroscopy,164 Raman spectroscopy,165 electron spin response spectrometry,166 microchemical crystal tests,167 168 ultraviolet spectroscopy/nuclear magnetic resonance/polarography.169... [Pg.114]

Ultraviolet-visible (UV-Vis) spectrophotometric detectors are used to monitor chromatographic separations. However, this type of detection offers very little specificity. Element specific detectors are much more useful and important. Atomic absorption spectrometry (AAS), inductively coupled plasma-atomic emission spectroscopy (ICPAES) and inductively coupled plasma-mass spectrometry (ICP-MS) are often used in current studies. The highest sensitivity is achieved by graphite furnace-AAS and ICP-MS. The former is used off-line while the latter is coupled to the chromatographic column and is used on-line . [Pg.403]

The first exposure to spectroscopy for most scientists is ultraviolet/ visible absorbance. As virtually every HPLC chromatograph employed in the pharmaceutical industry uses UV absorbance as the detection method, it is no wonder that the most popular hyphenated technique is HPLC-DAD. DAD spectrographs have been coupled to all liquid-based chromatographic systems including HPLC (preparative, analytical, and microbore), capillary electrophoresis (CE) and supercritical fluid chromatography (SFC). There have been several successes with TLC plates,18 but it is more common for developed plates to be scraped and the sample analyzed offline. [Pg.351]

The derivatization of analytes is very important in several branches of analytical chemistry. It expands the fields of application of various spectroscopic techniques (ultraviolet-visible (UV-vis), fluorimetry, nuclear magnetic resonance (NMR), and mass spectroscopies), and in several cases increases also the selectivity and sensitivity of these techniques. Derivatization is also an inevitable tool in all chromatographic and electrophoretic techniques. In gas chromatography (GC), the main importance of derivatization is the improvement of the volatility/thermal stability of the analytes, and in all of the discussed separation techniques it has the potential of increasing the selectivity of the separation (including enantiomeric separations) and the sensitivity of the detection. [Pg.841]
