One glycosylation site exists on the P suburhts of human LH [53664-53-2 and TSH [64365-92-OJ ie, Asn-30 (Fig. 4). In some species, Asn-13 of LH-P is glycosylated (48). FSH-P suburnt [58857-12-8] is glycosylated at two sites, Asn-13 and 30. Based on interactions of synthetic peptides with the LH receptor, loops formed by P93—100 and P38—57 may be essential for hormone bioactivity (48). Highly conserved sequences between residues 31—37 have been implicated in the formation of the a—P suburnt dimer (48), which is absolutely necessary for the expression of bioactivity. [Pg.177]

Thyroid-stimulating hormone can be used clinically to test thyroid function but has not found practical apphcation in the treatment of human thyroid insufficiency. Thyroid autonomy appears as a solitary toxic nodule or toxic multinodular goitre. In toxic thyroid, the nodule s synthesis and secretion of thyroid hormones is autonomous from the thyroid-stimulating hormone (TSH), which is produced in the pituitary gland. Accordingly TSH is suppressed and the extranodular thyroid tissue is functionally downregulated. Thyroid autonomy occurs frequently in iodine-deficient countries, whereas it is much less common in iodine-sufficient areas. Constitu-tively activating mutations in the TSH receptor and in the Gs a protein are the major molecular aetiology of toxic thyroid nodules. [Pg.1201]

Thyrotropin (TSH) regulates the production and secretion of thyroid hormones as well as thyroid epithelial cell growth via the TSH receptor. The TSH receptor belongs to the family of G protein-coupled receptors. It is composed of 764 amino acids.
