Triturates, tablet

Tablets, hypodermic Molded tablet triturates intended to be dissolved in water to make a solution to be injected parenterally. The usual weight of hypodermic tablets is about 0.03 g, which distinguishes them from ordinary tablet triturates that weigh about 0.06 g. Formerly prepared extemporaneously by pharmacists, modern compressed hypodermic tablets are not intended to be sterile, although they are manufactured under strict conditions as a precaution against contamination (see Triturates, tablet) ...

Triturates, tablet Small, disk-like masses of medicinal powders prepared by forcing a moistened tablet mass into a die by manual pressure and allowing them to dry and harden. The basis of tablet triturates is usually a mixture of lactose and sucrose in a 5 1 ratio, moistened with a volatile liquid such as alcohol. Tablet triturates were introduced in New York in 1878 by Dr. Robert W. Fuller as a palatable and convenient means of administering potent drugs by mouth. Fuller s original triturates consisted of triturations of metallic. 

The biotin market is divided between agricultural and human use, with —90% of biotin used in the animal health care market and —10% for the human nutritional market. The major producers of biotin are Hoffmann-La Roche, Lon2a, E. Merck-Darmstadt, Rhc ne-Poulenc, Sumitomo Pharmaceutical, E. Sung, and Tanabe Seiyaku (100). Worldwide production of biotin in 1994 was approximately 60 metric tons. The Hst price for pure biotin in 1995 was — 7.00/g whereas, the Hst price for technical feed-grade biotin was — 5.50/g. Biotin is used in various pharmaceutical, food, and special dietary products, including multivitamin preparations in Hquid, tablet, capsule, or powder forms. One of the commercially available products of i7-biotin is Britrit-1, which is a 1% biotin trituration used in food premixes. 

Tablets are solid dosage forms that are compressed or prepared by a sintering proeess, including sublingual, buccal, chewable, effervescent, and compressed tablets. Some of these ean be easily compoimded an example of a tablet triturate is as follows ...

As an example, ethinyl estradiol tablets are powdered and triturated with four 20-ml portions of chloroform, decanted, filtered, and analyzed by TLC [3, p. 639]. Numerous other examples can be found in the latter half of this book. 

Identification. Triturate a quantity of finely powdered tablets, containing the equivalent of about 100 mg of dipyridamole, with 10 mL of 0.1 N hydrochloric acid, and filter. Add 0.1 N sodium hydroxide to the filtrate until the solution is basic and a precipitate forms. Heat the mixture on a steam bath for 1 min, cool, filter, and dry the residue at 105°C for 1 h. The residue so obtained responds to the identification tests for dipyridamole. 

The factor by which the dissolution of an active substance is increased by triturating it with crospovidone usually lies between 2 and 10 but can exceed 10. This is shown in Fig. 82 for medroxyprogesterone acetate tablets, in which a six-fold excess of crospovidone improves the dissolution rate by a factor of 5, compared with that of tablets without this excipient. But in the some cases such factor of 5 already can be achieved by the ratio 1 + 2 (see Figs. 80 and 83).

LSD has been dispersed in numerous forms. Delysid (LSD-25) from Sandoz was distributed in ampules and also tablets containing 100 rg.. LSD that appeared outside of research circles was usually triturated (dosed) on sugar cubes and in tablet form (both as compressed tablets and molded tablets) of many sizes and colors. LSD that is dispersed into films (clearlight, window pane) of gelatin, agarose or cellulose also appears in a variety of colors and shapes. The most common form of LSD appears in blotter paper form.

Sample preparation Triturate 1 tablet with a glass rod with 5 mL water, sonicate for 20 min, extract with 9 mL dichloromethane then three times with 5 mL dichloromethane, filter (paper), make up to 25 mL with dichloromethane. Remove a 500 p,L aliquot, add 200 pL 1.2 mM phenacetin in dichloromethane -I- 100 pL 0.5 mM 4-dimethylaminopyri-dine + 100 pL 100 mM N-CBZ-Phe in dichloromethane -I- 100 pL 100 mM N, N -dicy-clohexylcarbodiimide in dichloromethane, shake mechanically at 30° for 1 h, inject a 10 pL aliquot.
