Ticarcillin pharmacokinetics

Pharmacokinetics Widely distributed. Protein binding ticarcillin 45%-60%, clavulanate 9%-30%. Minimally metabolized in the liver. Primarily excreted unchanged in urine. Removed by hemodialysis. Half-life 1-1.2 hr (increased in impaired renal function). 

The in vivo activity of a combination of clavulanic acid and amoxycillin is shown in Table 6.4 [30]. This -lactamase inhibitory activity, together with useful pharmacokinetic properties, has resulted in the development of Augmentin (potassium clavulanate in combination with amoxycillin) for oral and parenteral use and Timentin (potassium clavulanate plus ticarcillin) for parenteral use, against a wide range of bacterial infections. ...

Although there is limited pharmacokinetic information available in horses, these agents appear to be similar to other penicillins. Following i.v. injection of ticarcillin to horses, at a dose rate of 44 mg/kg, the serum concentration at 30 min was 104.3 jjig/1 and the mean peak peritoneal fluid concentration (61.4(i,g/l) was reached 2h after injection. The half-life of ticarcillin was 0.94 h. Following i.m. injection (44 mg/kg), the peak serum (28.3 xg/l) and peritoneal fluid (19.2 xg/l) concentrations were reached after 2h. The bioavailability of ticarcillin was 64.9%.

The drug possesses an almost similar antibaeterial profile and pharmacokinetic characteristics as those of carbenicillin (I). Because, of the following two positive advantages ticarcillin proved to be highly erucial and vital in the treatment of serious infeetions e.g., Ps aeruginosa) that may require a high-dose therapy ... 

No interaction was seen in 10 patients given tobramycin with piperacillin, or in another 10 healthy subjects given once daily gentamicin with piperacillin/tazobactam. Only minimal pharmacokinetic changes were seen in 9 healthy subjects given tobramycin with piperacillin/tazobactam, and 18 cystic fibrosis patients (adults and children) given tobramycin with ticarcillin."... 

The pharmacokinetic changes seen in the studies with cloxacillin and ticarcillin were not considered clinically relevant. In addition, the haemostatic changes seen in the study with cloxacillin were unlikely to he due to an interaction. The manufacturer notes that in clinical use danaparoid has frequently been used with a variety of drugs, including antihacterials, and that there is no evidence of any direct interaction with danaparoid. ... 

No interaction normalfy occurs between digoxin and amoxicillin, cefazolin, cefuroxime, flucloxacillin, phenoxymethylpenicillin or ticarcillin/clavulanic acid. No pharmacokinetic interaction occurs between ampicillin and digitoxin. In contrast, one early study found that cefradine increased serum levels of digoxin.
