Thioglycolate-induced peritonitis

X10 thioglycollate-induced peritoneal macrophages were incubated with arabinogalac-tan or medium for 18 h at 37°C in 5% C02- Supernatants were collected and serially diluted onto 2 X10 L929 cells treated with 2.5 pg of actinomycin/ml.

A -Tetrahydrocannabinol inhibited lipopolysaccharide- and IFN-y-induced nitric oxide production by thioglycollate-elicited peritoneal macrophages of female BALB/c mice (Coffey et al. 1996). 

Adherent, thioglycolate-induced, starch-induced peritoneal mouse or bone marrow macrophages are incubated for 24 h with the substance to be investigated. The supernatant of the culture is then discarded, the macrophages washed, and coincubated with the Cr labeled tumor cells for 24 h. 

Several patents claiming aryl sulfonamides have recently appeared [117-121], In the first, compound 43 was described to be effective in the rat collagen-induced arthritis model when dosed at 100 mpk (s.c., q.d., from day 9-17) [117]. This patent also reported that compound 44 significantly inhibited the number of monocytes recruited following thioglycolate injection in a rat peritonitis model. Two other patent applications describe sulfonamides 45 [120] and 46 [121] as CCR2 antagonists. 

Finally, routine animal models for inflammatory diseases can be used for testing the in vivo efficacy of selectin antagonists the murine peritonitis model and the ear edema model are the most common. In the murine peritonitis model [200], the migration of leukocytes in response to an acute inflammatory stimulus is assessed by intraperitoneal injection of thioglycolate. In the arachidonic acid- or croton oil-induced ear edema model [131,201], inflammation is measured as neutrophil infiltration, represented by myeloperoxidase activity in ear biopsy samples.
